CD27isamemberofthetumornecrosisfactor(TNF)receptorsuperfamily,whichincludestheTcellco-stimulatoryreceptorsOX40,4-1BBandherpesvirusentrymediator(HVEM).CD27isexpressedonvarioustypesofTcells,Bcellsandasubsetofnaturalkillercells.ItactivatesNF-BandMAPK/JNKsignalinguponinteractionwithitsTNF-likeligand,CD70,whichisexpressedbynumeroustumorcells.AdaptorproteinsTRAF2andTRAF5canalsostimulateCD27signaling.ActivationofCD27leadstolymphoidproliferation,differentiation,apoptosis,andtheinductionoflong-termmemory.TheCD27/CD70pathwayisakeytargetforthedevelopmentoftreatmentsforcancerandinflammatorydiseases.
