Description:
TheJARID1CHomogeneousAssayKitisdesignedtomeasureactivityoftheJARID1Cforscreeningandprofilingapplications,usingAlphaLISA®technology.ThekeytotheJARID1CHomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordemethylaseactivitydetection.First,asamplecontainingJARID1Cenzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:
AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
