TheFluorogenicSIRT2AssayKitisacompleteassaysystemdesignedtomeasureSirtuin2(SIRT2)activityforscreeningandprofilingapplications.Itcomesinaconvenient96-wellformat,withallthereagentsnecessaryfor32fluorescentSIRT2activitymeasurements*.ThekitincludespurifiedSIRT2enzymeandaSIRTinhibitor,Nicotinamide,foruseasapositiveandnegativecontrol,respectively.TheFluorogenicSIRT2AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheSIRT2activitylevel.First,theSIRTfluorometricsubstrate(SIRTsubstrate1),containinganacetylatedlysinesidechain,isincubatedwithpurifiedSIRT2enzyme.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
