Background:
HDACsregulatecellularprocessesbycatalyzingthehydrolysisofanacetylgroupfromacetyllysinesinmodifiedproteins.IntheHDACassay,fluorescent-dyemoleculesareattachedtoapeptidecontainingacetyllysine.Attachmenttothepeptidequenchesthefluorescenceofthedye.AftertreatmentofthepeptidewithanHDAC,thereactionismixedwithadevelopmentsolutionthatisspecificfornonacetylatedlysines.IftheacetylgrouphasbeenremovedfromthelysinebytheHDAC,thissolutionwillreleasethedyeallowingforfluorescence.FluorescenceisthereforedirectlyrelatedtoHDACactivity.
Description:
TheFluorogenicHDAC9AssayKitisacompleteassaysystemdesignedtomeasurehistonedeacetylase9(HDAC9)activityforscreeningandprofilingapplications.Itcomesinaconvenient96-wellformat,withallthereagentsnecessaryfor100fluorescentHDAC9activitymeasurements.Inaddition,thekitincludespurifiedHDAC9enzymeandapotentHDACinhibitor,TrichostatinA,foruseasapositiveandnegativecontrol.TheFluorogenicHDAC9AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheHDAC9activitylevel.First,theHDACfluorometricsubstrate,containinganacetylatedlysinesidechain,isincubatedwithpurifiedHDAC9.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
