Background:

HDACsregulatecellularprocessesbycatalyzingthehydrolysisofanacetylgroupfromacetyllysinesinmodifiedproteins.IntheHDACassay,fluorescent-dyemoleculesareattachedtoapeptidecontainingacetyllysine.Attachmenttothepeptidequenchesthefluorescenceofthedye.AftertreatmentofthepeptidewithanHDAC,thereactionismixedwithadevelopmentsolutionthatisspecificfornonacetylatedlysines.IftheacetylgrouphasbeenremovedfromthelysinebytheHDAC,thissolutionwillreleasethedyeallowingforfluorescence.FluorescenceisthereforedirectlyrelatedtoHDACactivity.

Description:

TheFluorogenicHDAC8AssayKitisacompleteassaysystemdesignedtomeasurehistonedeacetylase(HDAC)8activityforscreeningandprofilingapplications.Itcomesinaconvenient96-wellformat,withallthereagentsnecessaryfor100fluorescentHDACactivitymeasurements.Inaddition,thekitincludespurifiedHDAC8andapotentHDACinhibitor,TrichostatinA,foruseasapositiveandnegativecontrol.TheFluorogenicHDAC8AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwoasimplestepsonamicrotiterplateareneededtoanalyzetheHDAC8activitylevel.First,theHDACfluorometricsubstrateisincubatedwithpurifiedHDAC8enzyme.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
