Background:
HDACsregulatecellularprocessesbycatalyzingthehydrolysisofanacetylgroupfromacetyllysinesinmodifiedproteins.IntheHDACassay,fluorescent-dyemoleculesareattachedtoapeptidecontainingacetyllysine.Attachmenttothepeptidequenchesthefluorescenceofthedye.AftertreatmentofthepeptidewithanHDAC,thereactionismixedwithadevelopmentsolutionthatisspecificfornonacetylatedlysines.IftheacetylgrouphasbeenremovedfromthelysinebytheHDAC,thissolutionwillreleasethedyeallowingforfluorescence.FluorescenceisthereforedirectlyrelatedtoHDACactivity.
Description:
TheFluorogenicHDAC7AssayKitisacompleteassaysystemdesignedtomeasurehistonedeacetylase7(HDAC7)activityforscreeningandprofilingapplications.Itcomesinaconvenient96-wellformat,withallthereagentsnecessaryfor100fluorescentHDAC7activitymeasurements.Inaddition,thekitincludespurifiedHDAC7enzymeandapotentHDACinhibitor,TrichostatinA,foruseasapositiveandnegativecontrol.TheFluorogenicHDAC7AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheHDAC7activitylevel.First,theHDACfluorometricsubstrate,containinganacetylatedlysinesidechain,isincubatedwithpurifiedHDAC7.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
