TheSREReporterKitisdesignedformonitoringtheactivityoftheERKsignalingpathwayandthetranscriptionalactivityofSRFinculturedcells.Thekitcontainsatransfection-readySREluciferasereportervector,whichisanERKpathway-responsivereporter.ThisreportercontainsthefireflyluciferasegeneunderthecontrolofmultimerizedSREresponsiveelementslocatedupstreamofaminimalpromoter.TheSREreporterispremixedwithaconstitutively-expressingRenilla(seapansy)luciferasevectorthatservesasaninternalcontrolfortransfectionefficiency.Thekitalsoincludesanon-induciblefireflyluciferasevectorpremixedwithconstitutively-expressingRenillaluciferasevectorasanegativecontrol.Thenon-inducibleluciferasevectorcontainsthefireflyluciferasegeneunderthecontrolofaminimalpromoter,withoutanyadditionalresponseelements.Thenegativecontroliscriticalfordeterminingpathway-specificeffectsandthebackgroundluciferaseactivity.
