ThePDE4Dcell-basedactivityassayisdesignedforscreeninginhibitorsofPDE4D7inculturedcells.TheassayisbasedontransfectingcellswiththeCREluciferasereporter.CREreportercontainsthefireflyluciferasegeneunderthecontrolofcAMPresponseelement(CRE).ElevationofintracellularcAMPactivatesCREbindingprotein(CREB)tobindCREandinducetheexpressionofluciferase.WhencellstransientlytransfectedwithCREreporterareactivatedbyforskolin,theintracellularlevelofcAMPisupregulated,whichinducestheexpressionofCREluciferasereporter.However,whencellsareco-transfectedwithPDE4D7expressionvectorandCREreporter,thelevelofforskolin-inducedcAMPisreduced,resultinginlowerexpressionlevelofluciferase.WhencellsaretreatedwithPDE4DinhibitortoinhibitPDE4D7activity,cAMPlevelisrestored,resultinginhigherluciferaseactivity.
