Description:

TheNotchPathwayReporterkitisdesignedformonitoringtheactivityofNotchsignalingpathwayintheculturedcells.Thekitcontainsthetransfection-readyexpressionvectorforNotch1withtheentireextracellulardomaindeleted(Notch1DE).Insidethecells,theNotch1DEcanbecleavedby-secretaseandactiveNotch1NICDisreleasedtonucleus.ThekitalsocontainsaCSL(CBF1/RBP-J)luciferasereportervector,whichisaNotchpathway-responsivereporter.ThisreportercontainsafireflyluciferasegeneunderthecontrolofmultimerizedCSLresponsiveelementupstreamofaminimalpromoter.TheCSL(CBF1/RBP-J)reporterispremixedwithconstitutivelyexpressingRenilla(seapansy)luciferasevector,whichservesasaninternalpositivecontrolfortransfectionefficiency.

Thekitalsoincludesanon-inducIBLefireflyluciferasevectorpremixedwithconstitutivelyexpressingRenillaluciferasevectorasanegativecontrol.Thenon-inducibleluciferasevectorcontainsafireflyluciferasegeneunderthecontrolofaminimalpromoter,butwithoutanyadditionalresponseelements.Thenegativecontroliscriticalfordeterminingpathwayspecificeffectsandbackgroundluciferaseactivity.
