Background:

NF-κB(NuclearFactor-KappaB,NFkB)isaheterodimericproteincomposedofdifferentcombinationsofmembersoftheRelfamilyoftranscriptionfactors.TheNF-kB/Relfamilyoftranscriptionfactors(p50,p65,c-Rel,etc.)areinvolvedinstress,immune,andinflammatoryresponses.Inunstimulatedcells,theNF-κBdimersaresequesteredinthecytoplasmbyinhibitoryIkBproteins.Proinflammatorycytokines,LPS,growthfactors,andantigenreceptorsactivateIBkinase(IKK),whichphosphorylatestheIBproteins.PhosphorylationofIkBleadstoitsdegradation,freeingNF-κBcomplexestotranslocatetothenucleus,bindtoNF-κBDNAresponseelements,andinducethetranscriptionofthetargetgenes.

TheNF-κBReporterkitisdesignedformonitoringtheactivityoftheNF-κBsignalingpathwayintheculturedcells.Thekitcontainstransfection-readyNF-κBluciferasereportervector.ThisreportercontainsafireflyluciferasegeneunderthecontrolofmultimerizedNF-κBresponsiveelementlocatedupstreamofaminimalpromoter.TheNF-κBreporterispremixedwithconstitutively-expressingRenilla(seapansy)luciferasevector,whichservesasaninternalcontrolfortransfectionefficiency.Thekitalsoincludesanon-inducIBLefireflyluciferasevectorpremixedwithconstitutively-expressingRenillaluciferasevectorasanegativecontrol.Thenon-inducibleluciferasevectorcontainsafireflyluciferasegeneunderthecontrolofaminimalpromoter,withoutanyadditionalresponseelements.Thenegativecontroliscriticaltodeterminingpathwayspecificeffectsandbackgroundluciferaseactivity.
